JPMorgan Maintains "Neutral" Rating on BioNTech (BNTX.US): Bullish on the Potential of pumitamig Combined with B7-H3 ADC, but Key Data Readout Not Expected Until After 2028
JPMorgan maintains a "Neutral" rating on BioNTech with a price target of US$92.
The European Society for Medical Oncology (ESMO) Annual Congress was held in Madrid, Spain, from October 23 to 27. JPMorgan released a research report on BioNTech (BNTX.US) after attending the conference. JP Morgan stated its conclusion bluntly at the opening of the note: this batch of updates at ESMO, "while positive, does not materially change our thesis given the data sets remain early-to-mid stage and the timeline to key readoutsparticularly pumitamig out to 20282030+remains long." JP Morgan maintains a "Neutral" rating on BioNTech with a price target of US$92.
After communicating with the company following the conference, the bank confirmed that BioNTech is directing its efforts toward combination therapies, with pumitamig as the main backbone, and the typical combination being pumitamig + B7H3 ADC. The note then discussed several key presentations at the conference one by one.
Stepping back, JPMorgan's stance is wait-and-see: although a considerable portion of BioNTech's market capitalization is supported by its cash balance, it is in no rush to step in at current levels before pipeline visibility becomes clearer.
pumitamig: First Phase 2 data in second-line lung cancer, clearly superior to the docetaxel benchmark
ROSETTA Lung-107 is a global Phase 2 study evaluating pumitamig (a PD-L1VEGF-A bispecific immunomodulator, co-developed with Bristol-Myers Squibb Company) combined with docetaxel in patients with second-line non-small cell lung cancer (NSCLC) who progressed after immunotherapy + chemotherapy. Among efficacy-evaluable patients, the confirmed objective response rate (cORR) was 37% and median progression-free survival (mPFS) was 6.9 months (data cutoff July 10, 2026; median follow-up 12.2 weeks).
The benchmark for comparison is critical: historical expectations for second-line docetaxel monotherapy are approximately 13% ORR and 3.54 months mPFS. Responses were observed across all PD-L1 expression levels; broken down, squamous patients (n=7) had a cORR/unconfirmed ORR of 57%, while non-squamous patients (n=23) had 30%/39%.
On safety, JPMorgan considers the profile acceptable: Grade 3 or higher treatment-related adverse events (TRAEs) related to pumitamig were 18%, and 8% of patients discontinued due to pumitamig-related TRAEs; among adverse events of interest, all bleeding events (16%) were Grade 1, and no Grade 3 or higher hypertension or proteinuria events occurred.
The bank's conclusion is unsurprising"we are not surprised to see greater benefit with the addition of pumitamig on top of docetaxel"and it believes this second-line data set further supports the benefit of pumitamig in the global NSCLC population.
The timeline is a constraint repeatedly emphasized in this note: pumitamig is being tested in 4 global Phase 3 studiesfirst-line NSCLC (ROSETTA Lung-02, data expected 2029), first-line NSCLC with PD-L1>50% (ROSETTA Lung-202, 2030 and beyond), consolidation after chemoradiotherapy in unresectable Stage III NSCLC (ROSETTA Lung-201, 2030 and beyond), and first-line small cell lung cancer (ROSETTA Lung-01, 2028).
BNT324 (B7H3 ADC): Combination data more striking, 6mg/kg is the preferred dose
BNT324 is an antibody-drug conjugate (ADC) targeting B7-H3. The conference presented updated Phase 2 monotherapy data in platinum-resistant ovarian cancer (PROC), as well as the first data for its combination with pumitamig in PROC.
The combination regimen produced a striking number: cORR of 71% (including 2 responses pending confirmation), with median follow-up of 6.5 months (data cutoff September 4, 2026, n=31).
On monotherapy, the 6mg/kg every-three-weeks regimen is viewed by JPMorgan as the more favored dosebased on the company's communications with them and the fact that "this is the dose for which combination data with pumitamig exist." At this dose, 16 patients had a cORR of 50% (unconfirmed ORR 62.5%), median duration of response of 6.9 months, and mPFS of 8.3 months. The 9mg/kg every-three-weeks regimen showed higher efficacy metrics but clearly greater toxicityGrade 3 or higher TRAEs were 81%, versus 44% in the 6mg group.
The conclusion is: whether as monotherapy or in combination, BNT324 demonstrates promising characteristics; the monotherapy data are competitive, but after communicating with the company, JPMorgan senses that the priority for BNT324 lies in combination with pumitamig.
pumitamig+BNT324: Early activity across tumor types, liver cancer data most closely watched
This global Phase 2 trial presented the first results of pumitamig+BNT324 across multiple solid tumors, continuing the small cell lung cancer combination data previously presented at the World Conference on Lung Cancer (WCLC). The ESMO presentation added early antitumor activity in head and neck cancer, melanoma, liver cancer, cervical cancer, and platinum-resistant ovarian cancer.
Follow-up across the five tumor types ranged from 4.87.7 months; encouragingly, in patients previously treated with immunotherapy and/or Avastin (bevacizumab), unconfirmed ORR and confirmed ORR were similar to the overall population. The hepatocellular carcinoma (HCC) and platinum-resistant ovarian cancer data received particular attention in the presentation: among 21 HCC patients (median 2 prior lines of therapy, of whom 19 had prior IO/Avastin), the combination regimen's unconfirmed ORR/ORR was 43%/38%. The safety profile was manageable, with no dose-limiting toxicities (DLTs), and overall consistent with the established profiles of each agent.
gotistobart: Melanoma no longer a priority, focus shifts to Phase 3 head-to-head
BioNTech also presented longer follow-up data from a Phase 1/2 study of gotistobart combined with pembrolizumab in relapsed/refractory melanoma previously treated with CTLA-4 and PD-(L)1: the 3mg/kg group (n=22) had a cORR/mPFS of 18%/2.6 months, and the 6mg/kg group (n=27) had 22%/2.1 months.
Although ORR and mPFS were similar between the two groups, median overall survival was very noisy: 28 months in the 3mg/kg group (33 months of follow-up) versus 11 months in the 6mg/kg group (41 months of follow-up).
After communicating with the company, JPMorgan made clear: melanoma (post-IO treatment) is not the priority direction for gotistobart. The company emphasized the Phase 3 head-to-head studygotistobart monotherapy versus docetaxel in second-line squamous NSCLC, with overall survival as the primary endpoint and topline data expected this year.
Valuation and stance
This note did not adjust the rating or price target. JPMorgan maintains its "Neutral" rating and US$92 price target, implying approximately 5.9% upside from the current price of US$86.86on the more conservative end among institutions covering the stock. The bank's wait-and-see rationale is not directed at valuation itself but at timing: key readouts for the core assets are concentrated in 20282030, while current data are mostly early-to-mid-stage results from small samples with short follow-up.
JPMorgan also points out a structural fact: a considerable portion of BioNTech's market capitalization is supported by its cash balancemeaning there is some downside buffer, but also meaning the share price is more dependent on future pipeline delivery; until the data are clearer, the bank chooses not to rush in. It should be noted that this note does not separately list upside/downside risk factors, nor does it provide valuation methodology parameters (such as DCF, WACC, etc.); it is in the format of a conference note rather than a full rating report.
