Hutchison China MediTech Limited (00013) announced that the combination therapy of Savolitinib (ORPATHYS) and Osimertinib (TAGRISSO) has achieved positive topline results in the SAFFRON Phase III study compared to platinum-based doublet chemotherapy for the treatment of patients with non-small cell lung cancer (NSCLC) harboring epidermal growth factor receptor (EGFR) mutations. The results demonstrate a statistically and clinically significant improvement in progression-free survival (PFS) and overall survival (OS). The tumors of patients in the study exhibited high levels of MET overexpression or amplification, and they had previously experienced disease progression after treatment with TAGRISSO.

Third-generation EGFR tyrosine kinase inhibitors (TKIs) have significantly improved treatment outcomes for patients with EGFR-mutated NSCLC. However, one-third of patients may develop MET overexpression or amplification, which is one of the most common mechanisms leading to resistance after third-generation EGFR TKI treatment. MET-driven resistance is often associated with poorer prognosis, and there remains a significant unmet clinical need for effective and well-tolerated treatment options in the post-line therapy setting.
The safety profile of the combination therapy of Savolitinib and Osimertinib is consistent with the known safety profiles of each drug, and no new safety issues have been identified. The data will be presented at an upcoming academic conference and shared with global regulatory authorities. Based on the SAFFRON Phase III study, Savolitinib and Osimertinib combination therapy has been approved in China for the treatment of locally advanced or metastatic NSCLC with EGFR mutations and MET amplification after disease progression following EGFR TKI treatment. Savolitinib is co-developed by AstraZeneca and HUTCHMED and is commercialized by AstraZeneca.
